Starting my 4 stage RODI after a few years in storage. Changed all internals. Everything is hand tight. Carbon Block has a trickle leak right at the top, gave it another 1/8 turn and I’m still seeing the smallest of leaks. Is replacing an O-ring an option? I don’t want to over tighten.

I’d try unscrewing. Removing and cleaning O ring and then putting back in

As a mechanic I lube all rubber o-rings with food grade silicone grease (even on cars) as it doesn't eat away rubber the way petroleum based lubes do. Worth a shot before replacing the seal
